We had a car and drove. It allowed us to stop along the way to see things especially the library at the University of Coimbra. It has been called the most beautiful college library in the world. Certainly worth the stop.
Buses and trains ply this route numerous times per day. There is usually no need to book in advance unless you have a specific time you must depart. In this case, buying a ticket one day prior is fine. You can buy your tickets at the bus/train station.

We have always had a car hire in Portugal but you can easily travel by train, the journey is about 2.5 hours. Reasonable fare varies from 40 to 24 euros depending on type of train and class of ticket. Personally I would book if possible. Try the Trainline Europe website.
