You're 2 blocks from the tube (Victoria Station) which is 1 change + 1 stop from the Piccadilly station. Give it a leisurely 10 minutes from 41 to boarding, another 15 mins train to theater I'd expect. Not terrible, I'd be fine with my wife doing it in evening shoes without discomfort.

You could get the Heathrow express to Paddington and then get the underground to Victoria. The hotel is less than a 5 minute walk from Victoria. Or you could get an Uber from Paddington to the hotel. It would be about £15 for an executive car. You can get the underground to Wembley.

no... not on the property but they have a contract with a gym right around the corner. The gym has classes as well as most of the typical machines for working out.

Is there parking for cars?
While I am not positive, I am fairly certain that there is no parking available. 41 Hotel is across a busy thoroughfare from Buckingham Palace and it is not clear where there might be a garage. There are some garages in the area. The concierge is wonderfully attentive so I recommend contacting the Hotel directly.
