Either leave the car in Florence, or leave the car at Venezia Porto Marghera or Venezia Mestre. These are the train stations just outside of Venice. It really would be your only option to drive "to" Venice. You'd park and leave the car there, then hop on a train into Venice-proper (Stazione di Venezia Santa Lucia). The Marriott does have a boat go to the train station a few times a day, but your best bet would be to take the public water ferry (vaporetto) to San Marco, and get the Marriott shuttle ferry from there.
