It's quite easy to take the AirTrain and subway from JFK to this hotel. Catch the AirTrain to the subway line and get on the E train to Manhattan. Get off at Court Sq-23 St station, and choose your exit carefully so that you come out at 21st St: head toward the stairs you see as you get off the train. From there it's only a 5 min walk down 21st St to the hotel. From JFK this will take you a good hour and a bit. While this is the cheapest way to your hotel, and safe enough in my opinion, the quickest and easiest way is to catch a cab--but more $$$. Keep in mind the subway can get really full as you get closer to Manhattan, depending on what time you travel, so you don't want to have too much luggage with you.
To catch the subway into Manhattan, go back to this same station and catch the train you want going in that direction (the E line for example): it's only one stop away! (Depending where you're headed.)
Depending how long you're staying in NYC, you might want to buy a one-week MetroCard for unlimited subway travel--it's quite cheap, around $35. Even if you're staying less than a week, it doesn't take long for this to pay for itself. Otherwise you can buy a MetroCard and fill it with as much credit as you like so you don't need to continually buy subway tickets. Purchase your MetroCard at a vending machine as you're preparing to get on the E train from JFK to your hotel.
