All public transport options, I believe, are contained in this excellent effort -

Sorry, no comment about car rental.
Plenty of suggestions as well as a Forum for the Algarve on Trip Advisor:

Better rates obviously with the return to same place. Or Lisbon.
The road between the airport at Faro and Seville is good. Bear in mind that if you are staying in Seville old city you will need to find a suitable car park and this could cost say €15 per day or thereabouts. Some hotels will have spaces. I stay this month in Silken Al Andalus on the southern end of Seville city but within reach of the old city by bus or taxi in say 15 minutes and I think I got car parking in the hotel park for €6 . I say I think because have not got the confirmation with me ! Some car rental companies in Faro just do not like you taking their car to Spain and charge extra for the pleasure.

Just one thing though - http://www.lagoscarhire.com/ - does not do one way rentals Faro to Seville, one returns.
http://iberiacarhire.com/ is the only one that does one way.
Thanks so much for all of your help!
